摘要
目的探讨缺氧预处理及大鼠局灶性脑缺血后GFAP表达变化。方法建立大鼠缺氧预处理及局灶永久性脑缺血(PMCA)模型,应用免疫组化方法观察预缺氧(8%O2,92%N2,3h)+PMCAO不同时间(6h、1d、3d、7d)GFAP表达的变化。结果脑缺血后随缺血时间延长,GFAP表达逐渐增多,于缺血后7d达高峰。预缺氧(8%O2,92%N2,3h)+PMCAO组各时间点GFAP表达进一步增强,尤其在缺血周边区明显,与缺血对照组相比差异具有显著性(P<0·05)。结论缺氧预处理可以使星形胶质细胞激活并反应性增生,GFAP表达变化可能是脑缺氧耐受的脑保护机制之一。
Objective To study the expression change of GFAP after hypoxia preconditioning and focal cerebral isehemia. Methods The a hypoxia preconditioning model and PMCAO model were induced and the expression of GFAP was examined by immunohistoehemieal staining. Results After cerebral isehemia, the expression of GFAP gradually increased and peaked at 7 d after isehemia. In the hypoxia preconditioning 3 h + focal cerebral isehemia group, the expression of GFAP increased significantly in the infarct border compared with isehemia control group(PMCAO) ( P 〈 0.05). Conclusion The astroeyte was activated and proliferated after hypoxia preconditioning and focal isehemia. It can be one of the protective meehanismof hypoxia preconditioning on neuron and may be correlation to the up-regulation expression of GFAP.
