脂质体介导的hIGF-1质粒促周围神经再生的实验研究

摘要 目的研究周围神经损伤处通过脂质体介导的人胰岛素样生长因子-1(hIGF-1)质粒的转染,观察其对周围神经再生的作用。方法雄性Wister大鼠30只,分别造成坐骨神经挤压伤模型,随机分成两组。hIGF-1治疗组:挤压伤处神经外膜下注射脂质体介导hIGF-1质粒10μl。生理盐水(SAL)组:挤压伤处神经外膜下注射生理盐水10μl。在术后行坐骨神经功能指数(SFI),电生理学和小腿三头肌肌湿重检测。结果hIGF-1治疗组电生理学和小腿三头肌肌湿重检测结果明显优于生理盐水组,两组差异有显著性意义(P<0·05),并能改善功能指数。结论脂质体介导的hIGF-1质粒能明显促进大鼠周围神经再生和预防肌肉萎缩。 Objective To investigate the regenerative effect of lipnsome-mediated human Insulin-like growth factor-1 (hIGF-1) plasmid given locally at a time on the peripheral nerve after injury. Methods 30 Wister male rats with crush pattern of sciatic nerve were randomly divided into two groups. Group hIGF-1:10μl of lipesome-mediated hIGF-1 plnsmid was locally injected in sciatic epineuRium of the crush spot. Group normal saline(SAL) : 10μl of normal saline was only given in epineurium of the crush spot. Sciatic function index(SFI), electrophysiology and wet weight of muscles triceps surae were detected after surgery. Results Results of elec- trophysiology and wet weight of muscles triceps surae in group hIGF-1 were obviously outweight to group SAL with a significance of difference(P 〈 0.05), and SFI was also improved. Conclusion The liposome-mediated hIGF-1 plasmid can significantly promote peripheral nerve regeneration and provent musclar atrophy.
