冬虫夏草制剂对肾小球硬化大鼠细胞外基质成分和TIMP-1 mRNA表达影响的研究 被引量:13

摘要 目的观察虫草制剂对进行性肾小球硬化大鼠细胞外基质成分和基质金属蛋白酶组织抑制因子-1(TIMP-1)mRNA表达的影响。方法将正常Wistar大鼠36只随机分为正常对照组、模型组和治疗组。模型组、治疗组分别行5/6肾切除制备进行性肾小球硬化模型。治疗组每天给予虫草制剂(百令胶囊4g/kg.d)水溶液灌胃,进行干预治疗,模型组和正常对照组每天给予等量的自来水灌胃。末次术后4周、9周分别观察肾脏病理变化,用免疫组织化学检测肾组织胶原IV、纤维连接蛋白(FN),用RT-PCR方法检测肾脏基质金属蛋白酶组织抑制因子-1(TIMP-1)mRNA的表达。结果治疗组能降低肾小球硬化指数,减少胶原IV、FN表达;减少TIMP-1mRNA的表达。结论虫草制剂能够减少5/6肾切除大鼠肾脏胶原IV、FN表达;抑制TIMP-1mRNA的表达;减轻肾小球硬化细胞外基质的积聚。 Objective To investigate the effect of Cordyeepssinensis on the renal extraeellular matrix(ECM) of Wistar rat with progressive glomeruloselerosis on the renal and the level of tissue inhibitor of metalloproteinase-1 (TIMP-1) mRNA of wiast rat with progressive glomeruloselerosis in rats, Methods 36 healthy Wiast rats were equally divided into three groups including control group, progressive glomeruloselerosis group and Cordyeepssinensis-treated progressive glomeruloselerosis group, The seeond and the third groups were used to be the progressive glomenruloselerosis model by 5/6 kidney being nephreetomized. The treated group would be treated with Cordyeepssinensis, eontrary to the eontrol group and progressive glomeruloselerosis group would be treated with eaqually water. The renal pathological changes of three groups were investigated after 4 weeks and 9 weeks. Immunohistoehemistry was used to examine the expressions of fibroneetin (FN), collagen Ⅳ. RT-PCR was used to examine the mRNA of TIMP-1. Results The Cordyeepssinensis-treated progressive glomeruloselerosis group can reduce glomeruloselerosis index and the expressions of FN, eolla- gen Ⅳ ; can reduce the level of TIMP-1mRNA. Conclusion Cordyeepssinensis ean reduce the expressions of FN, eollagen Ⅳ ; and inhibit the level of TIMP-lmRNA. Cordyeepssinensis ean alleviate the accumulation of extraeellular matrix.
