内毒素对大鼠肝细胞线粒体膜电位的影响及黄芪注射液的干预作用 被引量:13

摘要 目的:观察内毒素血症对大鼠肝细胞线粒体膜电位(△Ψm)的影响及黄芪注射液的干预作用。方法:将40只大鼠随机分为正常对照组及内毒素注射后6h、12h组和黄芪处理组。分离肝细胞并检测其线粒体△Ψm水平,同时提取大鼠肝细胞线粒体测定其丙二醛(MDA)的含量及超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSHPX)的活性。结果:在注射内毒素6h后,线粒体△Ψm减少,MDA的含量升高,SOD、GSHPX活性减少,12h后,线粒体△Ψm明显减少,MDA升高更加明显,SOD、GSHPX活性明显减少。与对应的内毒素组比较,经黄芪处理后,肝细胞线粒体△Ψm升高,MDA的含量明显减少,SOD、GSHPX活性升高。结论:内毒素血症能造成肝细胞线粒体△Ψm下降和线粒体的氧化损伤,黄芪注射液可有效抑制内毒素血症肝细胞线粒体△Ψm的下降,减少内源性自由基的生成,提高机体的抗氧化损伤能力,具有保护肝细胞的作用。 Objective: To observe the effect of endotoxin on hepatocyte mitochondrial transmembrane potential (Δψm) in rats and the intervention action of Astragalus Membranacenus injection. Methods: Forty healthy adult rats were randomly divided into normal control group and 6, 12 hours groups after injecting endotoxin and Astragalus Membranacenus treatment group, then separating hepatocyte and measuring the level of hepatocyte mitochondrial Δψm, the contents of MDA in hepatocyte mitochondria, and the activitiea of mitochondrial SOD and GSH-PX at the same time.Results: After injecting the endotoxin 6 hours, mitochondrial Δψm was reduced, MDA contents gone up, SOD, GSH-PX activities were reduced, after 12 hours, mitochondrial Δψm was obviously reduced, MDA gone up more and obviously. SOD, GSH-PX activities were obviously reduced. Comparing endotoxin groups, the Astragalus Membranacenus treatment, hepatocyte mitochondrial Δψm was gone up obviously, MDA contents were obviously reduced, SOD, GSH-PX activities were gone up obviously. Conelusion: Endotoxemia can cause hepatocyte mitochondrial Δψm to descend and mitochondrial oxidative injury. The Astragalus Membranacenus injection can effectively repress the descent of endotoxemia hepatocyte mitochondrial Δψm due to endotoxemia, reduce the formation of endogenous free radicals and raise the ability to anti-oxidative injury ability, having the function of protecting the liver cell.
