食管鳞癌Skp2蛋白表达及与P27的相关性

摘要 目的研究Skp2和P27蛋白在食管鳞癌中表达,探讨它们之间的关系及其临床意义。方法应用免疫组化Sp法检测70例食管鳞癌中Skp2和P27蛋白的表达并分析相关性。结果 Skp2蛋白在癌旁正常组织、不典型增生及食管鳞癌组中阳性表达率分别为10.00%(2/20)、37.04%(10/27)、41.43% (29/70),后两者的阳性率显著高于前者(P<0.05)。P27在正常组织的阳性率为85.00%(17/20),高于食管鳞癌组48.57%(34/70),其差异有显著性(P<0.01)。Skp2蛋白在食管鳞癌组织中表达Ⅲ-Ⅳ期高于Ⅰ~Ⅱ期(P <0.05),且与淋巴结转移显著正相关。P27蛋白表达与食管癌淋巴结转移负相关,Skp2蛋白表达与P27表达呈负相关。Skp2表达阳性及P27表达阴性提示预后较差(5年累积生存率40.63%,P<0.05)。结论 Skp2 和P27蛋白表达的检测可能成为判断食管鳞癌预后的指标,指导患者的临床治疗,并为早期诊断食管鳞癌提供参考。 Objective To study the expression of Skp2 protein and its relationship with the expression of P27 protein in esophageal carcinoma. Methods The expression of Skp2 and P27 protein was detected by immunohistochemistry in tissues from 70 patients with esophageal carcinoma.All patients was followed up for 5 yesrs or more.Results The positive expression rate of Skp2 in cases with normal esophageal tissues,esophageal precancerous lesion and esophageal carcinoma were 10.00%(2/20), 37.04%(10/27) and 41.43%(29/70)respecdrely.There was significant difference between normal esophageal tissues and esophageal precancerous lesion or esophageal carcinoma(P〈0.05).The P27 labeling frequency in normal tissues85.00% (17/20) which was significantly higher than that in esophageal carcinoma48.57(34/70)(P〈0.01). The Skp2 protein in esophageal carcinoma tissue expressed remarkable Ⅲ-Ⅳ stage which is righer than Ⅰ-Ⅱ stage,and is positively correlated with lymphatic metastasis.The P27 protein expression was inversely correlated with lymphatic metastasis, Skp2 expression was inversely correlated with P27 in esophageal carcinoma.The Skp2 positive and P27 negative group revealed the worst survival rate among groups. Conclusion Expression of Skp2 and P27 may prove to be valuable in the treatment and prognosis assessment of esophageal carcinoma.
