枸杞多糖对染铅小鼠行为功能保护及脑细胞Fas抗原表达的影响 被引量:4

摘要 [目的]观察和探讨枸杞多糖对染铅小鼠行为功能保护作用。[方法]将小鼠分为5组,即空白对照组、染铅模型组、枸杞多糖分为高(3.6g/kg)、中(1.8g/kg)、低(0.9g/kg)剂量组。染铅各组于染铅后第2天灌服不同剂量枸杞多糖,分别于灌服构杞多糖后第3周和第6周用Morris水迷宫实验测试小鼠到达平台所需逃逸时间;实验第6周后取脑组织,用间接免疫荧光法测荧光指数。[结果]3个枸杞多糖剂量组均能明显影响小鼠到达平台逃逸时间,其中第3周逃逸时间为(23.76±4.77)^(24.67±3.66)s,而对照组为(16.90±3.91)s,模型组为(27.81±4.69)s;第6周,3个剂量组为(13.27±2.95)^(17.51±5.27)s,而对照组为(10.77±2.96)s,模型组为(18.99±6.01)s。枸杞多糖各剂量组明显抑制小鼠大脑皮质和海马区Fas抗原的表达,其中大脑皮质抗原表达率对照组(100.0±4.0)%,模型组(196.21±12.7)%,低剂量组(189.9±23.6)%,中剂量组(176.31±31.7)%,高剂量组(129.5±19.5)%;海马区抗原表达率对照组(100.0±4.0)%,模型组(189.1±16.2)%,低剂量组(177.5±22.6)%,中剂量组(162.3±19.5)%,高剂量组(137.1±26.2)%。[结论]枸杞多糖对改善脑功能记忆有明显作用,可推测枸杞多糖对脑细胞的保护机制可能是通过抑制Fas抗原表达而影响脑细胞凋亡实现的。 [ Objective ] To investigate and probe into the effect of Lycium barbarum polysaccharides on protecting brain function of lead-exposed mice. [ Method ] The mice were divided into five groups. The lead-exposed groups were fed with different dosage of LBP, since the next day, using Morris water maze the time taking to reach the hidden platform in three weeks and six weeks were measured separately.After the experiment, the brainwas taken to measur tanning indexes using indirect immunofluorencence. [ Results ] Different dosage of BLP can influence the refuge time markedly. In the third week, the refuge time in the control group was 16.90 ± 3.91s, in the model group 27.81 ± 4.69, in the high-dosage group to low-dosage group 23.76 ± 4.77-24.67 ± 13.66; In the sixth week: in the control group 10.77 ± 2.96, in the model group 18.99 ± 6.01, in the high- dosage group to low-dosage group 13.27 ± 2.95-17.51 ± 5.27 .High dosage group prevents the expression of Fas antigen in hippocampus and cortex obviously. In cortex the expression rate in the control group was 100.0 ± 4.0, in the model group 196.21 ± 12.7, in the low-dosage group 189.9 ± 23.6, in the mid-dosage group 176.31 ± 31.7, in the high-dosage group 129.5 ± 19.5; In hippocampus: in the control group 100.04± 4.0, in the model group 189.1 ± 16.2, in the low-dosage group 177.5 ± 22.6, in the mid-dosage group the function of memory was improved markedly, which can speculate that the function of 162.3 ± 19.5, and in the high-dosage group 137.1 ± 26.2. [ Conclusion ] LBP is able to protecting brain cells, it might implement by preventing expression of Fas antigen via influence apoptosis of cells.
