二氢卟吩e6光动力学疗法对人黑素瘤A-375细胞株的杀伤作用 被引量:3

摘要 目的探讨二氢卟吩e6(Ce6)光动力学疗法(Ce6-PDT)对体外培养人黑素瘤A-375细胞株的杀伤作用。方法将体外培养的人黑素瘤A-375细胞加入Ce6,浓度分别为2、4、7·5、15、30和60μmol/L,孵育12h,予波长652nm半导体激光照射,照射的功率密度为15·6mW/cm2,能量密度分别为2、5、10和20J/cm2,继续孵育12h,用四唑盐(MTT)比色法测定细胞存活率。结果(1)Ce6与细胞共同培养而不受光照的条件下对细胞的毒性较小,其浓度小于15μmol/L时各浓度组间细胞存活率差异无显著意义(P>0·05);浓度较高(>15μmol/L)对细胞的存活率影响较大(P<0·01)。(2)Ce6-PDT能有效地杀伤体外培养的人黑素瘤A-375细胞,其杀伤作用与Ce6浓度及光照剂量成正相关(P<0·01)。结论Ce6-PDT能有效地杀伤体外培养的人黑素瘤A-375细胞;Ce6在较大浓度下对细胞有毒性。 Objective To investigate the photodynamic effects of chlorine e6 (Ce6) on the killing ability to human malignant melanoma A-375 cell in vitro. Methods Cultured human malignant melanoma A-375 cells were incubated in a medium containing various concentrations(2,4,7.5, 15,30 and 60 μmol/L) of chlorin e6 for 12 h and illuminated with different light doses (2, 5, 10 and 20 J/cm2 ) using a semiconductor laser (wavelength 652 nm; energy density of 15.6 mW/cm^2 ). After the Ce6-PDT 12h the survival rate of human malignant melanoma A-375 cell was analyzed by MTT assay. Results ( 1 ) The cytotoxicity of Ce6 on human malignant melanoma A-375 cells was little while there was no irradiation and the concentrations of chlorin e6 were less than 15 μmol/L (P 〉 0.05). But if the concentrations of chlorin e6 were more than 15 μmol/L,even if there were no irradiation, it could affect the livability of human malignant melanoma A-375 cells (P 〈 0. 01 ). (2) Ce6-PDT can kill human malignant melanoma A-375 effectively in vitro. Higher phototoxicity was obtained with higher dye and/or laser doses. Among the various concentrations of chlorin e6 and laser doses, the livability of human malignant melanoma A-375 cells existed prominent discrepancy(P 〈0. 01 ). Conclusions Ce6-PDT can kill the human malignant melanoma A-375 cells effectively in vitro. When in high concentrations, Ce6 can do cytotoxicity on human malignant melanoma A-375.
