摘要
目的检测在缺氧条件下缺氧诱导因子2α对肝癌SMMC-7721细胞生长和侵袭能力的影响。方法缺氧培养条件下脂质体Lipofectamine介导缺氧诱导因子(HIF-20α)脱氧寡核苷酸转染SMMC-7721肝癌细胞,测转染前后HIF-2α mRNA量和蛋白量检验转染效果;MTY法测定细胞生长和黏附能力变化;Transwell小室测定细胞的体外侵袭能力。结果HIF-2αmRNA和蛋白表达在反义转染组显著低于正义组、随机组与对照组(P〈0.01)。HIF-2仪表达阻断对细胞生长和活力无显著性影响(P〉0.05),但显著抑制体外粘附能力(抑制率16.99%~31.07%)和侵袭能力(抑制率17.9%)(P〈0.01)。结论HIF-2α表达阻断可抑制SMMC-7721肝癌细胞的黏附和侵袭活性。对其生长无显著影响。
Objective To investigate the effects of hypoxia inducible factor- 2α (HIF -2α) on the proliferation and invasion of hepatoeellular carcinoma cell line SMMC -7721 under the anoxia condition. Methods Lipofeetamine - mediated HIF -2α oligodeoxynueleotide (ODN) was transfeeted into SMMC -7721 cells and the expressions of HIF- 2α mRNA and protein were detected to evaluate the transfeetion, The proliferative and adhesive activity was determined by methyl thiazolyl tetrazolium (MTF) method and the invasive activity was determined by a transwell cell culture chamber method. Results cantly by HIF- 2α antisense oligodeoxyribonueleotide (ASODN) ( P The expressions of HIF - 2α mRNA and protein were inhibited signifi- 〈0.01 ). The blockage of HIF- 2α expression significantly inhibited the adhesive activity ( inhibition ratio 16. 99% - 31.07% ) and invasive activity ( inhibition ratio 17.9% ) ( P 〈 0. 01 ), with no significant influence on the proliferative activity of SMMC - 7721 cells ( P 〉 0. 05 ). Conclusion The blockage of HIF - 2α expression inhibits the ad- hesive and invasive activity of SMMC -7721cells, and has no evident impact on their proliferation.
