摘要
目的探讨DPC4基因在非小细胞肺癌(NSCLC)中的表达及其与凋亡的关系。方法利用免疫组织化学S-P法检测52例NSCLC组织、19例相应癌旁正常肺组织中DPC4的表达,应用TUNEL技术对细胞凋亡情况进行了检测并计算凋亡指数。结果DPC4在肺癌原发灶中的阳性表达率为63.46%(33/52),与癌旁正常肺组织中的阳性表达率为89.47%(17/19)相比显著降低(P<0.05);DPC4与组织学类型、肿瘤细胞分化程度无关(P>0.05),但与淋巴结转移显著相关(P<0.05)。DPC4阳性组凋亡指数(AI)明显高于DPC4阴性组(P<0.05)。结论DPC4的低表达可能是肺癌发生过程的早期事件,可促进肺癌的淋巴结转移,DPC4的高表达可能促进细胞的凋亡。
Objective To study the expression of DPC4 (deleted in pancreatic carcinoma locus4, DPC4) in non - small cell lung carcinoma(NSCLC) and its relation with apotosis. Methods An immunohistochemistry S - P method was used to detect the expression of DPCA in NSCLC tissues from 52 patients with primary NSCLC and in adiacent normal lung tissues from 19 cases. The apotosis of NSCLC cells was detected by a TUNEL technique and the apoptotic index(AI) was calculated. Results The positive rate of DPCA was 63.46% (33/52) in primary lung cancer tissues, in comparison to the positive rate of 89.47% (17/19) in normal lung tissues, the DPC4 expression level decreased significantly and there was statistical significant difference ( P〈0.05). DPC4 was without assoeiation with tissue types and cellular differentiation ( P 〉0.05), but it was closely associated with lymph node metastasis ( P 〈0.05). The AI in TUNEL- positive group was higher than that in the TUNEL- negative group. Conclusion The low expression of DPCA gene can be an early event during lung carcinogenesis. Simultaneously, it promoted lymph node metastasis. The high expression of DPCA may promote the cell apotosis.
