中药复方热毒平提高中暑小鼠腹腔巨噬细胞吞噬能力的机制探讨 被引量:2

摘要 目的探讨中药复方制剂热毒平提高中暑内毒素血症小鼠腹腔巨噬细胞吞噬能力的机制,为该类药物在重症中暑治疗中的运用打下基础。方法在干球温度(34.5±0.5)℃,相对湿度(60±5)%的条件下建立小鼠中暑模型,将40只小鼠随机分为热毒平组、西黄芪胶组、生理盐水组、高温对照组和正常对照组。采血并提取小鼠腹腔巨噬细胞,用中性红法观察小鼠腹腔巨噬细胞的吞噬功能,流式细胞仪检测其凋亡情况,试剂盒检测血中肿瘤坏死因子-α(TNF-α)的值。结果热毒平组小鼠腹腔巨噬细胞的吞噬能力高于西黄芪胶组、高温对照组、生理盐水组(P<0.001),与正常组相近;凋亡百分率较其他三组有明显降低(P<0.001),与正常组相近;血中TNF-α的值低于西黄芪胶组、高温对照组、生理盐水组(P<0.001),与正常对照组相近。结论热毒平抗中暑内毒素血症的主要机制之一是在增强巨噬细胞吞噬能力,降低TNF-α的含量,减少巨噬细胞的凋亡中发挥重要作用。 Objective To investigate mechanism of traditional Chinese herb medicinal compound Reduping, a novel endotoxin-antagonist, enhancing phagocytosis of macrophages in abdominal cavities of mice with endotoxemia in heat stroke and provide scientific evidence for clinical application of such medicine in treatment for heat stroke. Method Animal model with heat stroke was established in mice under dry bulb temperature of (34.5 ± 0.5)℃ and relative humidity of (60± 5) %. Forty mice were randomly divided into five groups, including Reduping group, Xihuangqijiao group, group with normal saline, heat stroke control group and normal control group. Blood specimens were collected and macrophages were extracted from the abdominal cavities of mice. Phagocytosis of macrophages was evaluated by neutral red method, and its apoptosis was observed with flow cytometry. Plasma level of tumor necrosis factor-α (TNF-α) was measured by a special reagent kit. Result Phagocytosis of mice was significantly stronger in Reduping group than that in the other three groups ( P 〈 0.001), but similar to the control group. Percentage of apoptotic cells was significantly less in Reduping group than that in the other three groups ( P 〈 0.001), but similar to the normal control group. Plasma level of TNF-α in Reduping group was significantly lower than that in the other three groups ( P 〈 0.001), but similar to the normal control group. Condusion One of the most important mechanism of Reduping against endotoxemia in heat stroke is to enhance the phagocytic function of macrophages, but decreased plasma level of TNF-α and depression of apoptosis of macrophages probably also played an important role in it.
