乙型肝炎病毒preS2蛋白在Hela细胞中的表达

Expression of hepatitis B virus preS 2 protein in Hela cells

[目的]构建乙型肝炎病毒含3′末端缺失的preS/S基因表达载体并转染细胞,观察所构建载体在体外培养的真核细胞中的表达.[方法]采用基因重组技术构建含有CMV启动子及preS/S基因(adr亚型)开放阅读框的preS/S基因表达载体pcDNA 3.1-preS/St,并将其转染至Hela细胞,利用免疫细胞化学方法检测其表达情况.[结果]免疫细胞化学检测发现preS 2蛋白可在Hela细胞中表达.

OBJECTIVE Expression vector containing 3＇-truncated preS/S gene of hepatitis B virus was constructed and transfered to observe that expression vector would express in eucaryotic cell. METHODS Expression vector pcDNA 3.1-preS/St was constructed, containing CMV promoter and 3＇-truncated preS/S gene open reading frame by recombination DNA technique. It was identified by restriction enzyme digestion and confined by DNA direct sequencing. Hela cell was transfered with expression vector pcDNA 3.1-preS/St RESULTS The preS 2 protein was discovered in Hela cells with immune cytochemistry.
