Expression of Fas/Fasl Gene Protein and its Chinical Significance in Human Non-small Cell Lung Cancer

摘要 目的:研究Fas/FasL蛋白在非小细胞肺癌(NSCLC)的表达及其临床意义。方法:应用链霉菌抗生物素蛋白2过氧化物酶(SP)法染色技术测定65例NSCLC组织标本Fas/FasL蛋白的表达水平,并分析其与病理类型、区域淋巴结转移、病理分级及TNM分期之间的关系。结果:NSCLC组织中Fas蛋白表达率显著低于正常肺组织(46.15%vs80.00%,P<0.05),而FasL蛋白表达率显著高于正常肺组织(49.23%vs13.33%,P<0.05);FasL的表达与肺癌区域淋巴结转移有关,但Fas/FasL与肿瘤病理类型、病理分级及TNM分期无关。结论:Fas/FasL蛋白可能参与了非小细胞肺癌细胞凋亡的调节;检测FasL蛋白的表达有可能为临床判断NSCLC区域淋巴结转移、病变的发展以及评估预后提供依据。 Objective: To study the clinical significance of the expression of Fas/FasL gene protein in non-small cell lung cancer (NSCLC). Methods: By using SP immuno-histochemical method with anti-Fas and FasL multi2clonal antibody, we detected the expression of Fas/FasL gene protein in 65 cases of NSCLC and studied it s correlation with histological types, local lymph node metastases, pathological grades and TNM stages. Results: The positive expression rate of Fas (46.15%) in NSCLC was much lower than that (80.00%) in the normal lung tissues (P 〈 0. 05) ,but the expression rate of FasL (49.23%) in NSCLC was much higher than that ( 13.33% ) in the normal lung tissues (P 〈 0. 05 ). FasL was correlated with the stage of local lymph node metastases, but none of them had any correlation with histological types, patholo-gical grades or the TNM stages (P 〉 0. 05 ). Conclusions: The data demonst rated that Fas/FasL might involve in the regulation of apoptosis of NSCLC, and the expression of FasL might be used as an objective indicator of the metastatic potency of local lymph-node, extent of invation and the prognosis in NSCLC.
