摘要 建立了一种测定发酵液中香兰素及其副产物的反相高效液相色谱法。该法采用C18色谱柱,以甲醇、水、冰乙酸(体积比为40:60:0.5)的混合溶液作流动相,流速为1.0mL/min,紫外检测波长为280nm。发酵液中的各物质在一定范围内,其峰面积与质量浓度呈良好的线性关系,而且具有很好的精密度。香兰素的平均回收率为100.58%(n=3),RSD为0.54%。该方法能够快速、简便、准确地对发酵液中各种酚类物质进行定量分析。 An HPLC method for the determination of vanillin and other by - products in the fermentation broth is described. The separation was carried out on a versapack C18 column. The mobile phase of methanol:water: acetic acid (40: 60:0.5 [vol/vol/vol]) was pumped at a flow rate of 1.0mL/min. Sample detection was achieved at 280nm. Standard curve were established for phenolic compounds and external standards were used in quantifying biotransformation results. The average recovery of vanillin was 100.58% (n = 3), and its RSD was 0.54%. It is a rapid, sensitive and accurate analytical method.
