摘要
目的检测慢性粒细胞白血病(CML)患者外周血TCR Vβ2-、Vβ5-和Vβ17-Dβ1 sjTRECs的存在情况,从而了解相应3种Vβ亚家族naive T细胞的近期胸腺输出情况.方法利用半巢式PCR扩增9例CML患者外周血单个核细胞(PBMC)、CD4+细胞和CD8+细胞DNA的Vβ2-、Vβ5-和Vβ17-Dβ1sjTRECs,13例正常人外周血作为对照.结果TCR Vβ2-、Vβ5-和Vβ17-Dβ1 sjTRECs在部分正常人和1例CML患者CD4+或CD8+细胞可以检测到,而在CML患者PBMC中则未能检测到.结论在CML患者Vβ2-、Vβ5-和Vβ17-Dβ1 sjTRECs检出率低,与其胸腺输出相应的TCR Vβ亚家族naive T细胞水平低下有关.
Objective To analyze the TCR Vβ- ,Vβ5 - and Vβ17 - Dβ1 sjTRECs in peripheral blood of the patients with chronic myeloid leukemia (CML), evaluating the thymic recent output function of three Vβ subfamily naive T calls. Methods The Vβ2 - , Vβ5 - and Vβ17- Dβ1 sjTRECs were amplified in genomic DNA in peripheral blood mononuclear cells (PBMC), CD4^+ cells and CD8^+ cells from 9 cases with CML by semi - nested PCR. 13 normal individuals served as controls. Results TCR Vβ2 - ,Vβ5 - and Vβ17 - Dβ1 sjTRECs could be detected in PBMC from a part of normal individuals and one sample of CD4^+ cells or CD8^ + cells from CML cases, whereas Vβ2 - , Vβ5 - , Vβ17 - Dβ1 sjTRECs could not be detected in PBMC from patients with CML. Conclusions Low frequency of TCR V132 - ,Vβ - and Vβ17 - Dβ1 sjTRECs could be found in patients with CML, it may relate to the prominent reduction of thymic output function of corresponding TCR Vβsubfamily naive T cells.
