维生素D中毒剂量的动物实验研究

摘要 将兔分组肌注维生素D(VD)0.5万、1万、3万及6万IU/kg,观察血清25-羟维生素D(25-OHD)与血清钙含量的变化及给药后4周、12周肾脏和心血管的病理改变。结果显示:各实验组血清25-OHD给药后2~4周上升达高峰,与对照组有显著性差异;与用药剂量呈正相关。血清钙用药后2周各实验组皆升高,与对照组有显著性差异。病理:对照组和VD0.5万IU/kg组正常;1万、3万及6万IU/kg组部分兔有肾或主动脉肌层小灶性钙沉着。 In 1991, from August through November,we used rabbits as experimental animals,and intramuscularly injected vitamin D3 (VD3) 5, 000IU/kg,10,000IU/kg, 30, 000IU/kg and 60,000IU/kg in different groups,each group with 6 rabbits,and 4 rabbits as control group. Serum 25 hydroxycholecalciferol(25-OHD) and calcium were determined for each rabbit before and after medication. The animals were sacrificed 4 and 12 weeks after medication and pathological examina tions were performed to see the pathological changes in the kidneys and cardiovascular system. Be fore the experiments, mean serum 25-OHD and calcium content of each group was similar (P>0.05). One week after medication, the 25-OHD level of each experimental group elevated, and was the highest during second through fourth weeks, which was significantly difterent1 from the control group, and correlated with the dosage of VD3, which was maintained for 12 weeks.Two weeks af ter medication, serum calcium of the experimental groups elevated, especially in the VD3 30,000 IU/kg and 60,000IU/kg groups. No tissue calcification was found in the control and VD3 5,000 IU/kg groups;2 rabbits in VD3 10,000IU/kg groups 2 rabbits in 30,000IU/kg groups and 3 rab bits in 60,000IU/kg groups had spotted calcification in the kidneys, in some cases in the muscle of aorta. This study indicated that for rabbits the smallest toxic dosage of VD3 was 10,000IU/kg,and the largest safe dosage was 5,000IU/kg.
