IL-1β和TNF-α在小鼠巨细胞病毒性心肌炎组织中的表达及其意义

摘要 目的研究IL-1β和TNF-α在巨细胞病毒(M CM V)心肌炎小鼠心肌组织中的表达及在心肌损伤中的作用。方法将60只4周龄BALB/C小鼠随机分为两组,实验组(36只)腹腔注射10-4TC ID M CM V,对照组(24只)注射3T 3细胞裂解液。观察腹腔注射后3、7、14d心肌组织中IL-1βTNF-α的表达及心肌病理改变。结果实验组心肌组织出现灶性或弥散性炎性细胞浸润、心肌细胞变性和坏死,对照组无病理改变。实验组IL-1β、TNF-α蛋白主要表达在炎性浸润细胞、变性或坏死的心肌细胞,而对照组几乎无表达。结论IL-1β、TNF-α在M CM V心肌炎心肌组织中表达,并可能在其发病中起重要作用。 Objective: To study the expression of IL-1β and TNF-α in the myocardium of murine cytomegalovirus (MCMV) myocarditis and to evaluate its effect on myocardial damage of MCMV myocarditis. Methods: Sixty BALB/C mice of 4 weeks were randomly divided into two groups: 36 were injected intraperitoneally with MCMV (experimental group) and 24 served as control group. Immunohistochemistry technique was used to detect IL-β and TNF-α expression in myocardium, and myocardial lesions were observed by histopathology. Results: Focal or diffuse lesions characterized by inflammatory cells and degeneration or necrosis of myocytes in experimental group were revealed in the myocardium; Positive staining signals of IL-1β and TNF-α proteins which mainly located in infiltrating inflammatory cells and degenerative or necrotic myocytes were markedly detectable. There were no positive findings in control group. Conclusion: IL-1β and TNF-α can be found in myocardium of MCMV myocarditis model. IL-1β and TNF-α may play important roles in pathogenesis of viral myocarditis.
