摘要
对法夫酵母类胡萝卜素的光谱学特征、分光法测定法夫酵母类胡萝卜素总量、高效液相色谱测定法夫酵母虾青素进行了研究.在二甲基亚砜丙酮混合液中,法夫酵母类胡萝卜素最大吸收峰的波长范围为473~481 nm;以虾青素为标准样,用分光法测定法夫酵母类胡萝卜素总量和用高效液相色谱法测定法夫酵母虾青素标准曲线回归模型的R2值超过了0.999,样品的回收率分别在98.85%~102.60%和99.96%~102.44%之间,RSD值分别在0.30%~7.00%和0.61%~2.26%之间,都在定量分析的阈值范围内,能满足定量分析的要求.用建立的方法对分批培养的法夫酵母样品进行分析,结果表明,法夫酵母主要在对数生长期和稳定期合成类胡萝卜素.
Spectrum characteristic of carotenoids of Phaffia rhodozyma, determination of total carotenoids by spectrophotometric method and determination astaxanthin by high performance liquid chromatography method were studied. Dissolved in dimethylsulpboxide (DMSO) and acetone mixture in ratio of 1:2, highest absorbency of carotenoids of Phaffia rhodozyma and astaxanthin were both occurred in 473~481 nm. it was showed that R^2 value of regressive model of calibration curve for determination total carotenoids and astaxanthin were both in excess of 0.999, recoveries were in range of 98.85%~102.60% and 99.96%~102.44%, RSD values ranged from 0.30% to 7.00% and 0.61% from to 2.26%, which all indicated the proposed method was accurate and decisive for quantitative analysis. Results of carotenoid analysis during fed-batch cultivation demonstrated that Phaffia rhodozyma synthesized carotenoids in exponential stage and stationary stage.
