摘要
目的:探讨一些因素对体外培养骨髓基质细胞增殖和分化的影响。方法:实验于2001-01/2004-12在吉林大学中日联谊医院骨科完成。取四五月龄新西兰大白兔20只,体外培养骨髓基质细胞。将传代培养的第2代骨髓间充质细胞按1×104/孔的细胞密度培养24h后,更换培养液,同时添加不同浓度的碱性成纤维细胞生长因子、肝细胞生长因子,胰岛素、氢化可的松、地塞米松培养24h,应用噻唑蓝法检测上述不同细胞因子和激素对骨髓基质细胞的增殖作用,以Bessey方法测定碱性磷酸酶活性。结果:①成纤维细胞生长因子、胰岛素及氢化可的松能够明显地刺激骨髓基质细胞的增殖。②肝细胞生长因子对骨髓基质的增殖作用不十分明显。③地塞米松表现出明显的抑制骨髓基质细胞增殖却促进其碱性磷酸酶合成的作用。结论:一些细胞因子和激素能够明显影响骨髓基质细胞体外的增殖和分化。
AIM: To investigate the influences of some factors on the proliferation and differentiation of bone marrow stromal cells(BMSCs) cultured in vitro.
METHODS: The experiment was conducted in the Department of Orthopedics, China-Japan Friendship Hospital of Jilin University from January 2001 to Decender 2004. BMSCs isolated from 20 white New Zealand rabbits of 4-5 months old were prepared for in vitro culture. The cultured BMSCs of the second generation were cultured at a density of 1 ×10^4 per well for 24 hours; then, the culture medium was changed, and the cells were cultured in the medium containing basic fibroblast growth factor, liver cell growth factor, insulin, hydrocortisone and decacortin of different concentrations respectively for 24 hours. MTr method was performed to detect the roles of different cytokines and hormones in the proliferation of BMSCs. Bessey method was used to measure ability of alkaline phosphatase.
RESULTS: ①Fibroblast growth factor, insulin and hydrocortisone could obviously stimulate the proliferation of BMSCs. ②Effect of liver cell growth factor on the proliferation of BMSCs was not remarkable. ③Decacortin presented a remarkable effect on the inhibition of BMSCs proliferation; however, it had a marked role in accelerating the synthesis of alkaline phosphatase.
CONCLUSION: Some cytokines and hormones can influence the proliferation and differentiation of BMSCs cultured in vitro.
