摘要
目的:观察骨髓间充质干细胞体外培养中雌激素受体与雄激素受体的表达情况。方法:实验于2004-03/2005-01在广州中医药大学免疫研究室内完成。从SD大鼠骨髓中分离培养间充质干细胞,流式细胞仪鉴定,免疫细胞化学方法检测雌激素受体与雄激素受体的表达情况。结果:①流式细胞仪结果显示,各阳性细胞占总细胞数的比例分别为:CD34(0.34%),CD45(0.22%),CD11b(19.40%),CD80(0.44%),CD86(1.79%)总体表达为阴性,CD29(98.24%),CD44(97.49%),CD166(97.28%)总体表达为阳性,且此结果显示细胞的纯度很高,显示此细胞为骨髓间充质干细胞。②部分间充质干细胞呈雌激素受体阳性、雄激素受体阳性。雌激素受体阳性细胞占总细胞比率约为(23.80±3.37)%,雄激素受体阳性细胞占总细胞比率约为(31.27±2.28)%。结论:雌雄激素可直接通过其受体作用于间充质干细胞。雌雄激素减少所致间充质干细胞的异常分化,可能是原发性骨质疏松的发病原因之一。
AIM: To research the expression of estrogen receptor and androgen receptor of mesenchymal stem cells (MSC) cultured in vitro. METHODS: The experiment was conducted at the Immunologic Research Room of Guangzhou University of Traditional Chinese Medicine between March 2004 and January 2005. The MSC were separated from bone marrow of SD rats, and then were detected with flow cytometer. The expression of estrogen receptor and androgen receptor was detected with immunocytocbemical methods.
RESULTS: ①Results of flow cytometer showed that the rates of each positive cells in total number of cells were CD34(0.34% ),CD45 (0.22%), CD 11 b ( 19.40% ), CD80 (0.44%), CD86 ( 1.79% ) (collectivity was negative), CD29 (98.24%) ,CD44 (97.49%) ,CD166 (97.28%) (collectivity was positive), respectively. Besides, this result revealed that the purity of cells was high, and the cell was MSC. ② Part of MSC expressed positive estrogen receptor and positive androgen receptor. The ratio of positive estrogen receptor .occupied about (23.80±3.37)%, and the ratio of positive androgen receptor accounted for about (31.27±2.28)% of the total cells.
CONCLUSION: The estrogen and androgen can have effect on MSC directly by their special receptors. The reducing of estrogen and androgen cause the abnormal differentiation of MSC, which can be one of reasons for the episode of primary osteoporosis.
