青蒿素对小鼠迟发型超敏反应的免疫抑制作用 被引量:6

摘要 目的:探讨天然植物提取物青蒿素对小鼠迟发型超敏反应(delayed-type hypersensitivity response,DTH)的免疫抑制作用及可能的作用机制.方法:建立小鼠DTH模型,灌胃给予不同浓度的青蒿素、FK506及青蒿素与FK506联合用药,观测其对小鼠DTH的影响.结果:青蒿素各剂量组[除50 mg/(kg.d)组]、FK506组及FK506与青蒿素联合用药组与溶媒组比较,溶媒组小鼠的耳片肿胀程度、单核细胞浸润程度、细胞因子干扰素-γ(interferon-gamma,IFN-γ)及细胞间粘附分子1(intercellular adhesion molecule 1,I-CAM-1)的表达水平均明显增高,具有统计学意义(P<0.05).青蒿素在一定剂量范围内有一定的量效关系.结论:当剂量在150 mg/(kg.d)以上时,青蒿素对小鼠DTH反应具有免疫抑制作用,其作用在一定剂量范围内随剂量加大而增强,同时,青蒿素与FK506联合用药具有协同作用,其可能机制之一是通过抑制IFN-γ的产生和ICAM-1的表达从而抑制免疫反应程度. Objective: To investigate the immunosuppressive effect and related mechanism of Qinghaosu. Methods: A modle of delayed - type hypersensitivity response (DTH) induced by dinitrofluorobenzene (DNFB) in mouse was established and the effect of various concentrations of Qinghaosu, FKS06, FKS06 combined with Qinghaosu on DTH response were observed. Resuls: Except Qinghaosu 50mg/ (kd·d) group, The murine auricle weight difference of left and right 24 hours after stimulation, the infiltration of mononuclear cell, the level of Interferon - gamma ( IFN - γ) in serum and the expression of intercellular adhesion molecule 1 (ICAM- 1) were significantly decreased in the other groups of different concentrations of Qinghaosu, and the effect of Qinghaosu was related to the dosage is above 150mg/ (kg·d) and is dose dependent within a certain dosage range, it can also be find that Qinghaosu has a synergitic action with FK506. One of the mechanisms might be by inhibiting the production of IFN- γ in serum and downregulating the expression of local ICAM- 1 in the ear.
