石英晶体微天平对微生物的测定

摘要 基于石英晶体微天平(QCM)对粘度和密度的响应,利用涂有固体培养基薄膜的QCM通过对加入菌液后明胶培养基膜溶解后凝固过程的测定而间接测定微生物。在频率响应曲线中,频率开始上升时间是一个尖锐折点(TT),易准确测定。在1.0×102~4.1×107cels/mL范围内TT与表皮葡萄球菌的初始浓度的对数值线性相关,相关系数为0.98。此种测定方法更加快速(少于5h)且只需要微量的样品。 Based on the response of quartz crystal microbalance to viscosity and density, a QCM sensor coated with thin solid gelatine medium film was developed. After the inoculation of bacterium suspension the film dissolved and then coagulated gradually. The coagulating process of gelatine was monitored and the bacterium could be determined indirectly. In the frequency curve, the time at which the frequency began to increase was defined as TT and was a sharp turning point. Log C was linearly related to TT wthen the initial number of bacterium was in the range of 1.0×10 2~4.1×10 7 cells/mL, the correlation coefficient is 0.98. This method was rapid (the detection time was less than 5h) and only micro sample was needed.
