摘要
对薄层色谱法测定辅酶Q0的操作条件进行探索。选择了以丙酮为溶剂对样品进行溶解,以石油醚-乙酸乙酯(体积比9:4)为展开剂上行展开,密封展板后在波长269 nm下进行测量。其定性检测结果:各组分分离良好,斑点集中且清晰。定量检测结果:线性范围为0.4-6 μg,线性方程为Y(μv)=10195.6+30544.6X(μg)(n=6),相关系数为0.9983,加标回收率94.6%,仪器精密度RSD=0.07%(n=3),同板精密度RSD=0.79%(n=3)。
In this paper, the condition of thin-layer chromatographic determination of ubiquinone-0 is improved. We select solvent with acetone, the developing agent with petroleum ether and ethyl acetate (volume ratio: 9:4), closure the thin board and determine by 269 nm. The effect of qualitative determine is the group object separated completely, clear and concentrated spot on the plate were obtained. The effect of quantitative determination: range of linearity: 0.4 - 6tLg, regression equation : Y ( μV ) = 10195.6 + 30544.6X (μg) ( n = 6 ), r = 0. 9983, average recovery: 94.6 %, precision of instrument : average RSD = 0.07 % ( n = 3), precision on the same board : average RSD = 0.79 % (n =3).
