Selenium Distribution in Se- Enriched Ganoderma Lucidum

摘要 为深化富硒灵芝的基础研究,指导富硒灵芝的生产与应用,以不同含硒量的富硒灵芝子实体为试样,系统地研究了硒在富硒灵芝中的分布,结果表明:富硒灵芝子实体中81.71%~87.92%的硒是以有机硒形式存在,并主要与子实体中的蛋白结合(蛋白硒占总硒的56.0%~61.2%);水溶性蛋白和碱溶性蛋白是灵芝结合硒的主要蛋白,总结合硒量占蛋白硒的75.2%~79.2%。按照结合硒量的多少,硒在蛋白中的分布依次为:水溶性蛋白>碱溶性蛋白>醇溶性蛋白>盐溶性蛋白。此外,应用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)分离富硒灵芝子实体中的含硒蛋白组分,并采用氢化物原子荧光法(HGAFS)对蛋白或多肽条带按分子质量段切割进行硒含量测定,结果表明:在分辨出的13条蛋白条带中,分子质量在8.709~142.53kDa的蛋白或多肽均结合了不同程度的硒,但硒主要分布在分子质量不超过16kDa的蛋白或多肽中。 The distribution of selenium in Se-enriched Ganoderma lucidum (Se-GL) was investigated using Se-GL of different Se content as tested samples in order to promote both the research and application work in Se-GL. The results showed that 81.71-87.92% of total Se in Se-GL were in the organic form, and mainly associated with the protein in GL Se bound to the protein extracts accounted for 56.0%-61.2% of total Se. Moreover, 75.2%-79.5% of Se bound to protein was incorporated with water-soluble and alkaline-soluble protein extracts. The amount of Se existed in the different protein fractions decreased in the order of water-soluble, alkaline-soluble, ethanol-soluble, and sodium chloride-soluble protein extracts. Additionally, selenoproteins in Se-GL were also separated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and analyzed Se content using atomic fluorescence spectrophoteometry (HGAFS). 13 protein or polypeptide bands with molecular weight from 8.709 kDa to 142.53 kDa were identified. They all combined different amount of selenium. However, selenium was mainly distributed in the proteins or polypeptides with molecular weight of no more than 16 kDa.
