HPLC determination of protoberberines in Futai soft capsules

目的:建立同时测定妇泰软胶囊中小檗碱型生物碱HPLC定量法.方法:采用高效液相色谱法.以SHIMADZU VP-ODS(250 mm×4.6 mm,5 μm)为色谱柱;流动相:0.05 mol·L-1磷酸二氢钠溶液(用磷酸调节pH值至3)-乙腈(70:30);流速:1.0 mL·min-1;检测波长:265 nm;柱温:40℃;结果:盐酸药根碱、盐酸巴马汀、盐酸小檗碱分别在0.02～0.10μg ,0.05～0.26μg,0.03～0.16μg范围内.浓度与峰面积线性关系良好(r=0.999 9,0.999 8,0.999 7).平均回收率分别为:99.5%,101.3%,100.6%.结论:方法简便、快速准确、灵敏度高,重现性好.可作为妇泰软胶囊的含量测定法.

Objective： To establish a HPLC method for the determination of protoberberines in compound Futai soil capsules. Methods： The HPLC condition was as follows-SHIMADZU VP - ODS column （250 mm×4.6mm,5μm）with the column temperature of 40℃; an eluate composed of 0.05 mol·L^-1 NaH3PO4 （pH = 3 adjusted with H3 PO4 ） ： acetonitrile（70 ： 30） ; flow rate at 1.0mL·min^-1 ; and detection UV at 265 nm. Results： The calibrated linear curves of jatrorrhizine hydroehlolide, palmatine hydroehlolide and berbeline hydroehloride concentrations were within the range of 0.02- 0. 10, 0.05 -0.26 and 0.03 -0.16 μg, respectively. The correlation coefficients （r） were 0.999 9,0.999 8,0.999 7, respectively. The mean recovery were 99.5% （ RSD = 1.2% ）, 101 .3% （ RSD = 2.7% ） and 100.6% （ RSD = 1.2 ）, respectively. Conclusion：The sensitive and reliable HPLC method is good for the quantification of components in Futai soft capsules.
