摘要
本试验改进了肌肉中肌苷酸测定的高效液相色谱法。采用WatersAtlantisTMdC18色谱柱(5μm,Φ4.6mm×150mm),以50mmol/LpH6.5甲酸铵缓冲液(含5%甲醇)为流动相,流速1ml/min;柱温:25℃;紫外检测波长为254nm。结果表明,肌肉中肌苷酸含量测定结果的相对标准偏差为1.11%,回收率为100.33%~101.67%。该方法可更准确的应用于生鲜肉类食品的肌苷酸含量的检测分析。
Abstract : The HPLC assay of inosinic acid in animal muscles was improved in this experiment. The separation was carded out with Waters Atlantis^TM dC18 analytical column(5 μm, Ф4.6mm ×150mm). The mobile phase consisted of 50mmol/L pH6.5 ammonium formate buffer and methanol (95:5, V/V). The flow rate was 1 ml/min and UV detection was monitored at 254nm. The HPLC was performanced at 25 ℃. The results showed that the relative standard deviation of inosinic acid in muscle was 1.11% and its recovery was 100.33%- 101.67%. This method could be applied to assay inosinic acid content in raw meat food more accurately.
