摘要
目的:制备榄香烯脂质体瘤苗,观察对抗小鼠H22肝癌作用,寻找有效的抗肝癌生物疗法。方法:用改进的机械分散法制备β-榄香烯修饰脂质体瘤苗,使磷脂酰胆碱∶胆固醇∶十八烷氨∶β-榄香烯摩尔比为4∶4∶1∶1.5,并通过腹腔接种将脂质体包裹的H22抗原(LA g组)及脂质体共包裹β-榄香烯与H22抗原的制剂(LEA g组)分别免疫荷瘤小鼠,观察小鼠的生存期,M TT法测定淋巴细胞对H22肝癌细胞的杀伤作用。结果:LEA g组的生存期和淋巴细胞毒活性均优于LA g组(P<0.05)。结论:β-榄香烯可增强脂质体瘤苗的抗肝癌免疫效应。
Objective:To observe the antitumor effect of liposomal tumor vaccine modified by β-elemene and look for the effective tumor vaccine formulation. Methods: MLVs (multilamellar vesicles) containing β-elemene and H22 Ags were prepared by improved mechanical dispersion with the lipid and β-elemene molar ratio : PC(phosphal tidylcholine) : CH(cholesterol) : SA (stearylamine) : β-elemene = 4 : 4 : 1 : 1.5. H22 Ag entrapped in liposomes (LAg group),liposome coentrapped with H22 Ag and β-elemene(LEAg group)were inoculated intraperitoneally into BalB/c mice loaded with H22 Hepatoma cells, respectively;the survival period were compared,the lymphocyte cytotoxicity was valued by MTT assay. Results :The survival period was longer and lymphocyte cytotoxicity was stronger in LEAg group compared with that control (P〈0.05). Conclusions:β-elemene can enhance the antitumor immune response of liposomal tumor vaccine.
