L-Glu促进纯化培养的海马和皮质星形胶质细胞增殖的研究

摘要 目的研究L-Glu对纯化培养的皮质和海马星形胶质细胞促增殖作用的机制。方法将纯化培养的星形胶质细胞分为7组:①无血清培养基组,②含L-Glu(1mmol/L)的无血清培养基组,③含MCCG(100μmol/L)的无血清培养基组,④含MPEP(100μmol/L)的无血清培养基组,⑤含L-Glu+MCCG(分别为1mmol/L,100μmol/L)的无血清培养基组,⑥含L-Glu+MPEP(分别为1mmol/L,100μmol/L)的无血清培养基组,⑦含L-Glu+MCCG+MPEP(分别为1mmol/L,100μmol/L及100μmol/L)的无血清培养基组。流式细胞仪检测细胞周期的变化。结果L-Glu促进纯化培养的星形胶质细胞的增殖,单独加入mGluRs的竞争性拮抗剂MCCG或MPEP则L-Glu的促增殖作用减弱,同时加入两种拮抗剂则L-Glu的促增殖作用消失。结论L-Glu通过作用于mGluR3和mGluR5促进星形胶质细胞的增殖,两者可能在星形胶质细胞增殖方面具有协同作用。 Objective To investigate the mechanism of the proliferation-promoting effcet of L-glutamate (L-Glu) on the purified cultured hippocampal and cerebral astrocytes. Methods Purified cultured astrocytes were randomly divided into 7 groUps: serum free medium (control), serum free medium containing L-Glu (lmmol/L), serum free medium containing MCCG (100/.tmol/L), serum free medium contai- ning MPEP (100μmol/L), serum free medium containing L-Glu+MCCG (1mmol/L, 100μmol/L, respectively), serum free medium containing L-Glu+MPEP (1mmol/L, 100μmol/L, respectively) and serum free medium containing L-Glu+MCCG+MPEP (1mmol/L, 100μmol/L and 100μmol/L respectively) group. The variations in the cell cycle of astrocytes were measured by means of flowcytometry. Results L-Glu promoted the proliferation of astrocytes at the concentration of 1mmol/L. Each of the two antagonists of mGluRs, MCCG or MPEP, could decrease the promotion effect of L-Glu, an combined use of both antagonists could completely inhibit the proliferation of astrocytes. Conclusion The promoting effect of L-Glu on the proliferation of purified cultured hippocampal and cerebral astrocytes might be through the two types of mGluRs, mGluR3 and mGluRS, which probably cooperate in this process.
