清热解毒剂对小鼠免疫功能的调节 被引量:2

摘要 目的:观察经灌胃给予清热解毒颗粒后小鼠免疫功能的变化情况。方法:实验于2004-03/07在泰山医学院分析实验室完成。选择昆明种小鼠70只,随机分为7组,即清热解毒颗粒3.0g/kg,1.5g/kg,0.75g/kg组、免疫低下模型高、中剂量组、免疫低下模型对照组及空白对照组,每组10只。清热解毒颗粒3.0g/kg,1.5g/kg,0.75g/kg组、免疫低下模型高、中剂量组小鼠分别给予10mL/kg清热解毒颗粒3.0g/kg,1.5g/kg,0.75g/kg,3.0g/kg,1.5g/kg灌胃,1次/d,连续14d,空白对照组每日给予等量的生理盐水。免疫低下模型高、中剂量组及免疫低下模型对照组小鼠于给药后第3,3,5天腹腔注射75mg/kg环磷酰胺1次。通过巨噬细胞吞噬功能试验、定量溶血分光光度计测定法、淋巴细胞转化试验,观察不同剂量的清热解毒颗粒对小鼠免疫功能的影响。结果:纳入小鼠70只,全部进入结果分析,无脱失。①各组小鼠的巨噬细胞吞噬率比较:清热解毒颗粒3.0g/kg,1.5g/kg,0.75g/kg组小鼠的巨噬细胞吞噬率与空白对照组相近(P>0.05)。免疫低下模型高、中剂量组小鼠的巨噬细胞吞噬率均显著高于免疫低下模型对照组犤(29.90±13.62)%,(23.67±10.62)%,(10.00±4.22)%(t=22.01,15.30,P<0.01,0.05)犦。②各组小鼠血清溶血素水平(吸光度413)的比较:清热解毒颗粒3.0g/kg,1.5g/kg,0.75g/kg组小鼠的吸光度413均显著高于空白对照组(t=21.92,22.13,23.02,P<0.01)。免疫低下模型高、中剂量组小鼠的吸光度413均显著高于免疫低下模型对照组(t=22.96,23.14,P<0.01)。③各组小鼠脾淋巴细胞转化率(吸光度570~630)的比较:清热解毒颗粒3.0g/kg,1.5g/kg,0.75g/kg组小鼠的吸光度570~630与空白对照组比较,差异无显著性意义(P>0.05)。免疫低下模型高、中剂量组小鼠的吸光度570~630均显著高于免疫低下模型对照组(t=21.98,22.33,P<0.01)。结论:不同剂量的清热解毒颗粒能完全或部分对抗免疫抑制剂的抑制作用,对小鼠细胞和体液免疫均有一定保护和促进作用。 AIM: To explore the changes of immunological function in mice after being treated with Qingre Jiedu granules by gastric perfusion. METHODS: The experiment was conducted at the laboratory of analysis of Taishan Medical College between March and July 2004. Seventy Kunming mice were selected and assigned randomly into 7 groups: 3.0 g/kg, 1.5 g/kg, 0.75 g/kg Qingre Jiedu granule group, high, middle dose low-immunity model group, low-immunity model control group and blank control group with 10 in each group. The mice in the 3.0 g/kg, 1.5 g/kg, 0.75 g/kg Qingre Jiedu granule group and high, middle dose low-immunity model group were given 10 mL/kg Qingre Jiedu granules 3.0 g/kg, 1.5 g/kg,0.75 g/kg, 3.0 g/kg, 1.5 g/kg, respectively with gastric perfusion, once a day for 14 days. Those in the blank control group were given saline at the same volume every day. After administration, at the 3^rd, 3^rd and 5^th days the mice in the high, middle dose low-immunity model group, low-immunity model control group were treated with 75 mg/kg cyclophosphamide once through intrapefitoneal injection. Phagocytosis function trial on histocytes, quantitative hemolysis spectrophotometer determination method and lymphocyte transformation test were used to observe the effects of Qingre Jiedu granules at different dosage on immunological function in mice. RESULTS: Seventy included mice were all involved in the result analysis, without drop-out. (1) Comparison of phagocytosis rate of. histocytes in mice of every group: The phagocytosis rate of histocytes in mice of 3.0 g/kg, 1.5 g/kg,0.75 g/kg Qingre Jiedu granules group was close to that in the blank control group (P 〉 0.05). It was higher significantly in the high, middle dose low-immunity model group than that in the low-immunity model control group [(29.90±13.62)%, (23.67±10.62)%, (10.00±4.22)% (t=22.01,15.30,P 〈 0.01,0.05)]. (2) Comparison of serum hemolysin level in mice of every group (absorbance413)(A413): The A413 of mice in the 3,0 g/kg, 1.5 g/kg,0.75 g/kg Qingre Jiedu granules group was higher remarkably than that in the blank control group (t=21.92,22.13,23.02,P 〈 0.01 ). The A413 was higher markedly in the high, middle dose low-immunity model group than that in the low-immunity model control group (t =22.96,23.14, P 〈 0.01 ). (3) Comparison of transformation efficiency of lymphocytes of spleen in mice of every group (A 570-630): The A570-630 in mice of 3.0 g/kg, 1.5 g/kg, 0.75 g/kg Qingre Jiedu granules group had insignificant difference as compared with blank control group (P 〉 0.05). The A 570-630 in mice of high, middle dose low-immunity model group was higher obviously than that in the low-immunity model control group (t =21.98,22.33,P 〈 0.01 ). CONCLUSION: The Qingre Jiedu granules at different dosage can fully or partly confront inhibition of immuno-suppressive drugs, which have certain protective and accelerative effects both on cells and humoral immunity in mice.
