摘要 目的:通过分析细胞凋亡的信号转导途径,探讨半胱氨酸天冬氨酸蛋白酶家族与心血管疾病的关系。资料来源:应用计算机检索美国NCBIPubmed数据库1980-01/200402关于细胞凋亡、半胱氨酸天冬氨酸蛋白酶与心血管疾病的文章,主要检索主题词包括“caspases,cardivasculardiseases,apoptosis”,语言种类为English。资料选择:选取心血管疾病中,心肌细胞凋亡的信号转导途径的原创实验性文章以及2001年以来在权威期刊上发表的相关综述文章。纳入标准:①具有原创性,论点论据可靠的实验文章。②观点明确,分析全面的综述文章。③文献主体内容与本课题联系紧密的文章。排除标准:具有明显实验设计和结果错误的文章及观点模糊的综述。质量评价主要考察资料的真实性、实施过程是否严密。资料提炼:共检索到关于半胱氨酸天冬氨酸蛋白酶家族与心血管疾病的文献32篇,20篇文献符合纳入标准。排除的12篇文献均为重复性文章。资料综合:20篇文献分别阐明了在心血管疾病,如心肌梗死,心力衰竭高血压,肺动脉高压等的发生发展过程中,半胱氨酸天冬氨酸蛋白酶家族介导的凋亡发挥了重要的作用。应用半胱氨酸天冬氨酸蛋白酶抑制剂治疗心血管疾病的实验研究,已取得一定效果。结论:半胱氨酸天冬氨酸蛋白酶家族介导的凋亡,在多种心血管疾病的发生发展中,发挥着不可替代的作用,通过抑制其活性来对抗心肌细胞凋亡,从而保护心脏,有可能成为治疗心血管疾病的一项新策略。 OBJECTIVE: To investigate the association between caspases family and cardiovascular diseases through analyzing signal transduction pathway of cell apoptosis. DATA SOURCES: We searched the American NCBI Pubmed database for articles on apoptosis, caspases and cardiovascular diseases published between January 1980 and February 2004 with the key words of “caspases,cardivascular diseases,apoptosis”in English. STUDY SELECTION: The original experimental articles, which reported signal transduction pathway of myocardial cell apoptosis of cardiovascular disease and reviews about the topic published in authorized journals since 2001, were selected. Inclusion criteria: (1)Origin and reliable experimental articles; (2) Well analyzed review articles with clear opinions; (3) Articles with close relationship with the topic in this paper. Exclusion criteria: Articles with obvious errors in the experimental design or results and reviews with obscure opinions. Quality evaluation mainly inspected the validity of data and whether the experimental duration was accurate or not. DATA EXTRACTION: Totally 32 articles about family of caspases and cardiovascular diseases were collected. Twenty literatures were accorded with the inclusion criteria. The 12 excluded ones were repetitive. DATA SYNTHESIS:The 20 articles elucidated that family of caspases mediated apoptosis played an important role on the occurrence and development of cardiovascular diseases, such as myocardial infarction, cardiac failure, hypertension and pulmonary hypertension, etc. The experiment of using caspase inhibitor on the treatment of cardiovascular disease had gained certain effects. CONCLUSION: The apoptosis mediated by family of caspases plays an irreplaceable role in the occurrence and development of various cardiovasular diseases, and restrainning its activity to confront myocardial cell apoptosis so as to protect heart probably be a new method for cardiovascular disease.
