摘要
目的研究第三代双膦酸盐类药物阿仑膦酸钠(alendronate,固邦)对体外培养的破骨细胞的作用。方法建立骨质疏松大鼠模型,于0、2、4、8w进行体外骨髓破骨细胞样细胞(OLC)的培养,并进行阿仑膦酸钠干预,观察OLC数量和形态的变化。结果OVX组大鼠OLC数量高于C组和Sham组;在所有组中,阿仑膦酸钠均能使OLC显著减少(P<0.01)。结论大鼠去卵巢后OLC形成增加;阿仑膦酸钠能显著抑制OVX大鼠体外培养的OLC的形成。
Objective To investigate the effect of alendronate on in vitro osteoclast-like cell(OLC). Methods Sixty 2.5 to 3-month-old female Wistar rats were divided into three groups. Rats in group OVX were subjected to be ovariectomized, while rats in Sham group subjected to sham-operation. At the 0 week,2 weeks,4 weeks and 8 weeks after ovariectomy, femur bone marrow were cultured in the presence of macrophage colony stimulating factor and receptor activator of nuclear factor kappa-B ligand(RANKL).Alendronate was added to the culture medium to the final concentrations of 10-7 mol/L. Results The number of OLCs in OVX group were the highest among the three groups( P 〈 0.01 ). The number of OLCs in each group was greatly inhibited by alendronate ( P 〈 0.01 ). Conclusions Ovariectomization increases the derivation of bone marrow cells to OLCs which can be inhibited by alendronate.
