摘要 目的探讨蛋白激酶C(PKC)在单次异氟醚(Iso)预处理诱导的快速相脑缺血耐受中的作用。方法利用大鼠大脑中动脉阻闭模型(MCAO),观察Iso预处理前给予PKC拮抗剂chelerythrine(Che,侧脑室注射)对Iso预处理脑保护作用的影响。结果再灌注24h时脑梗死容积Iso组(139.6±46.0mm3)较空白对照组(264.3±62.7mm3)明显缩小(P=0.000),且明显小于Iso+Che组(286.4±96.0mm3)(P=0.000)和单纯Che组(273.8±69.5mm3)(P=0.000)。Iso+Che组、单纯Che组和空白对照组无明显差异。Iso+溶剂组(192.1±80.3mm3,P=0.108)、假手术组(197.1±66.6mm3,P=0.079)和Iso组相比无统计学差异。再灌注24h时神经功能障碍评分Iso组与空白对照组有降低的趋势,但无统计学差异(P>0.05)。结论PKC激活参与了单次Iso预处理诱导的快速相脑缺血耐受的形成。 Objective To investigate the role of protein kinase C (PKC) in acute phase of cerebra ischemic tolerance induced by isoflurane (Iso) preconditioning in rats. Methods Chelerythrine (Che), a specific PKC inhibitor, was intracerebroventricularly administered before lso preconditioning. And its influence on the neuroprotective effect of Iso preconditioning in the middle cerebral artery occlusion (MCAO) rats was observed. Results The infarct volume in Iso goup was significantly smaller than those in control (264. 3 ± 62.7 mmS)(P=0.000), Iso + Che(286.4±96.0 ramS) (P =0.000) and Che (273.8±69.5 mm^3) (P =0.000) groups, Tbere was no statistical difference among Iso + Che, Che and control groups ( P 〉 0. 05 ). There was no statistical differences among lso + vehicle, sham, and Iso groups (P 〉 0. 05). The neurologic defter scores in the Iso group showed a tendency to decease compared with the control, but there was no statistical difference among all the groups. Conclusion The activatian of PKC plays an important role in acute phase of cerebral ischemic tolerance induced by Iso preconditioning.
