模拟高原急性缺氧对大鼠下丘脑神经肽Y受体Y1 mRNA表达的影响

摘要 目的探讨高原急性缺氧对大鼠食欲及其下丘脑神经肽Y受体Y1mRNA表达的影响。方法雄性Wistar大鼠随机分为平原对照组、3000m缺氧组、4000m缺氧组,5000m缺氧组和6000m缺氧组,各缺氧组设置缺氧后1、2、3d3个时相点,观察低氧环境下大鼠食欲的变化;采用RT-PCR方法检测大鼠下丘脑中Y1受体mRNA的表达。结果①高原急性缺氧后,大鼠食欲减退。②对照组和缺氧组大鼠下丘脑中均存在Y1受体mRNA表达。高原急性缺氧后,下丘脑Y1受体mRNA表达降低,并随着海拔高度的上升和缺氧时间的延长逐渐降低。结论高原急性缺氧对大鼠食欲有抑制作用;高原急性缺氧后下丘脑Y1受体mRNA表达降低,这可能是大鼠食欲减退的重要原因之一。 Objective To explore the effect of acute hypoxia on the appetite and expression of Y1R mRNA in rat hypothalamus. Methods A total of 117 male Wistar rats were randomly divided to raise at plain (n = 9) , or in the altitude chambers imitating the environment at 3 000 m, 4 000, 5 000 m and 6 000 m for respectively 1 d, 2 d and 3 d (n = 9 at each altitude for different period of time). The change of appetite was observed and the expression of Y1R mRNA in rat hypothalamus was measured by using the technique of semiquantitative reverse transcription-polymerase chain reaction. Results After acute exposure to hypoxia, the appetite of rats decreased. The expression of Y1R mRNA could be detected in rat hypothalamus of hypoxia groups and control group. After acute exposure to hypoxia, the expression of Y1R mRNA decreased in rat hypothalamus with the rise of altitude and time prolonging (P 〈 0.05, P 〈 0.01 ). Conclusion The acute hypoxia might decrease food intake of rats, After acute exposure to hypoxia, the expression of Y1R mRNA decreased in rat hypothalamus, which is one of important reasons for loss of appetite.
