HB-PDT对经顺铂诱导耐药及非耐药肺癌细胞系杀伤效应的比较 被引量:1

Phototoxic effect of HB-PDT on DDP-resistant and non-DDP-resistant lung cancer cell lines:a comparative study

摘要 目的:对比研究耐药及非耐药肺癌细胞株对竹红菌乙素-光动力疗法(HB-PDT)的反应性,以及耐药肿瘤对HB-PDT及化疗药物顺铂的反应性之间是否存在差别。方法:以不同浓度的竹红菌乙素(HB)分别孵育人肺腺癌细胞系A549及经顺铂(DDP)诱导耐药的多药耐药细胞系A549/DDP,然后分别在铜蒸汽激光混合光饱和光剂量条件下进行照光处理,照光后继续孵育24 h,MTT法分别测定不同HB浓度下2种细胞的存活率,分别绘制HB对2种细胞存活率曲线并拟合曲线方程,根据方程求出HB对2种细胞的半数杀伤浓度(IC50)。结果:HB-PDT对经顺铂诱导耐药及非耐药细胞系均具有很强的杀伤效应,HB-PDT对A549及A549/DDP的IC50分别为33.82 ng/ml和34.19 ng/ml,两者之间差异不显著(P>0.05);顺铂对A549及A549/DDP的IC50分别为1.06μg/ml和8.06μg/ml,两者之间差异显著(P<0.01);HB-PDT和化疗药物顺铂对耐药肿瘤细胞杀伤效应间差异显著(P<0.01)。结论:HB-PDT对于耐药及非耐药细胞的光毒效应无显著差别。HB-PDT对耐药肿瘤细胞具有明显的光毒效应,其杀伤经顺铂耐药肿瘤细胞的效果明显优于已产生耐药的化疗药物。HB-PDT有可能发展为对顺铂耐药肿瘤的有效治疗手段。 Objective:To compare the reactivity of multidrug resistant and non-drug-resistant lung cancer cell lines to hypocrellin B and photodynamic therapy( HB-PDT), and to study whether there is difference in the reactivities of multidrug resistant lung cancer cell line to HB-PDT and chemotherapy. Methods: Human lung adenocarcinoma cell line A549 and DDP-induced multidrug-resistant cell line A549/DDP were separately incubated with different concentrations of HB,then irradiated by copper vapor laser with mixed wavelength light under saturated light dose. The survival rates of cells were measured by MTT assay 24 h after incubation. Accordingly cell survival curves of the 2 cell lines were drawn,the equations were derived and IC50 of each cell line was calculated. Results: HB-PDT had strong phototoxic effect on both DDP-resistant and non DDP-resistant lung cancer cell lines. The IC50 values of HB to A549 and A549/DDP were 33.82 ng/ml and 34. 19 ng/ml,respectively(P〉0.05) ; the IC50 values of DDP to A549 and A549/DDP were 1.06μg/ml and 8.06μg/ml,respectively(P〈0.01). HB-PDT and DDP had significantly different killing effects on multidrug-resistant A549/DDP cells(P〈0.01). Conclusion: The phototoxic effects of HB-PDT on DDP-resistant lung cancer cell lines have no significant difference. HB-PDT has strong phototoxic effect on DDP-resistant and non-DDP-resistant lung cancer cell line and its killing effect is superior to that of DDP. The phototoxic effects of HB- PDT to DDP-resistant and non-DDP-resistant lung cancer cell line are not significantly different. HB-PDT may become an effective therapy for DDP-resistant lung cancer.
