丁酸钠诱导Raji细胞表达DAPK的研究 被引量:7

摘要 目的探讨丁酸钠(SB)对人淋巴瘤Raji细胞生长和DAPK表达变化的影响。方法用丁酸钠作用于Raji细胞株,观察形态、增殖情况、细胞周期分布变化、死亡相关蛋白激酶(DAPK)基因启动子甲基化的变化及该酶表达的变化。结果成团悬浮生长的Raji细胞呈现贴壁生长,细胞增殖速度明显被抑制,细胞阻滞在G0/G1期;DAPK启动子去甲基化,DAPK表达量增加,细胞凋亡增加。结论丁酸钠具有诱导Raji细胞DAPK基因启动子去甲基化,使DAPK重新表达,诱导细胞凋亡的作用。 Aim To investigate the effect of sodium butyrate on Raji cells growth and the change of deathassociated protein kinase (DAPK) expression in Raji cells. Methods Raji cells were treated with sodium butyrate. Morphology of Raji cells was observed with microscope and electronic microscope. Cell growth was detected with MTT methods. Cell cycle was analyzed with flow cytometry. The change of methylation of DAPK gene promoter and the expression of DAPK were analyzed with methylation special PCR and RT-PCR.Results Raji cells were abserved in clusters suspending in the media or adhering to the wall. Cell growth was inhibited significantly. Cell cycle was arrested at G0/G1 phase. The DAPK gene promoter was demethylated and the expression of DAPK increased. Then cells were induced to apoptosis. Conclusion Sodium butyrate can induce demethylation of DAPK gene promoter resulting in DAPK expression and Raji cell apoptosis.
