摘要 目的研究ACE2在SARS-CoV感染中的作用。方法利用表达人类ACE2的小鼠成纤维细胞系和ACE2基因敲除小鼠研究SARS-CoV在小鼠成纤维细胞中的复制和ACE基因剔除小鼠体内的复制。结果SARS-CoV可以感染,并在表达人类小鼠ACE2的小鼠成纤维细胞中的复制,ACE2基因的缺失严重影响了SARS病毒与组织的结合,和SARS病毒在动物体内的复制,ACE2基因剔除对小鼠SARS-CoV感染具有保护作用。结论ACE2是SARS-CoV的主要受体,并且是SARS治疗潜在的靶基因。 Objective ACE2 is a receptor of SARS-CoV. In this paper, the replication of SARS-CoV in murine fibroblasts expressing human ACE2 and ACE2 knockout mice was studied. Methods The murine fibroblasts expressing human ACE2 and ACE2 knockout mice were used to investigate the replication of SARS-CoV and protective effect of ACE2 knockout on SARS infection in mice. Results The SARS-CoV infected and replicated in the murine fibroblast cells expressing human ACE2. The Block of ACE2 expression inhibited the infection and replication of SARS-CoV in vivo. The ACE2 gene knockout showed a protective effect on SARS-CoV infection in mice. Conclusion Our results suggested that ACE2 is a functional receptor for SARS- CoV and a possible target gene for SARS therapy.
