摘要
目的探讨豚鼠图形视网膜电图(patem electroretinogram,PERG)的记录方法和波形特点,为今后在视觉电生理研究中的应用提供依据.方法采用RETI-port系统,氯化银环状角膜电极为记录电极,自制不锈钢针状电极为参考电极置于记录眼颞侧皮下.846合剂麻醉后于明适应状态下记录成年豚鼠不同参数情况下的PERG.结果成年豚鼠PERG波形趋势较为明显,N1波在25 ms左右,P1波在47 ms左右,而N2波在94 ms左右,与人类较为相似,潜伏期较稳定;P1~N2幅值为2~6μV;随着空间频率的增加(0.12~0.02 cpd),幅值随之降低,幅值与空间频率呈负相关,当空间频率0.02~0.01 cpd时,幅值变化不明显;随着时间频率(刺激频率)的增大(2~8 Hz),幅值略有降低;随着对比度的增加(30%~97%),幅值随之增加,幅值与对比度呈正相关;随着平均亮度的增加,幅值随之增加,幅值与平均亮度呈正相关.结论成年豚鼠PERG波形以及变化趋势与人类较为相似,记录条件较为稳定,可以为模型动物应用于临床视觉电生理的相关实验.
Objective To investigate recording condition and characteristics of pattern electroretinogram (PERG) in guinea pigs, inorder to provide objective basis for applications of guinea pigs in visual electrophysiology. Methods Guinea pigs were kept on a special platform, which could be moved in three dimensions,and the distance between the tested eye and CRT were controlled by a self-made apparatus. A silver chloride loop electrode was contacted with corneal limbus as recording electrode. A stainless steel needle electrode was inserted in the temporal subcutaneous as reference. After guinea pigs were anaesthetized,PERG was recorded by RETI-port system. Results Waves of pattern electroretinograms in normal guinea pigs were clear and similar to those of humans. N1 wave located on 25 ms;P1 wave located on 47 ms and N2 located on 94 ms. Conclusion Waves and changing trends of PERG in normal guinea pigs are similar to those of humans. The recording condition is stable. As a result,guinea pigs can be used in clinical electrophysiology of vision.
