川芎嗪对内毒素刺激大鼠肝星状细胞增殖及瘦素和胶原表达的影响 被引量:9

摘要 目的观察川芎嗪对内毒素刺激肝星状细胞(HSC)增殖及瘦素表达的影响,并观察I型胶原表达的变化。研究川芎嗪抗肝纤维化过程中的机制。方法内毒素刺激大鼠肝星状细胞的基础上,加川芎嗪分组培养。MTT法检测其吸光度,ELISA法检测其上清液瘦素水平和I型胶原。结果内毒素刺激HSC增殖,川芎嗪抑制内毒素诱导的HSC增殖,并抑制I型胶原表达和瘦素的表达。结论川芎嗪抑制内毒素诱导的HSC活化和I型胶原表达。川芎嗪有可能通过抑制HSC增殖和抑制瘦素的分泌发挥抗纤维化作用。 Objective To observe the effects of tetramethylpyrazine(TMP) on endotoxin stimulate the proliferation of HSC and the expression of type Ⅰ collagen and leptin, to investigate the effect of TMP on against hepatic fibrosis Methods Rat HSC was stimulate with endotoxin and cultured with TMP, the cultured HSC were divided into groupes. Proliferation of rat HSC was detected with MTT technique. Expression levers of collagen Ⅰ and leptin was detected by Enzyme-Linked lmmunosorbent Assay (ELISA). Results Endotoxin significantly promoted proliferation of HSC ,. the proliferation of HSC and the increased expression levels of collagen Ⅰ and leptin induced by endotoxin could be inhibited by TMP. Conclusions TMP can inhibited the activiation and the expression levels of collagen 1 of HSC induced by endotoxin. TMP against hepatic fibrosis may be through the way of reducing the proliferation of HSC and the expression levers of leptin.
