摘要
目的观察糖尿病大鼠心肌病变过程中是否存在心肌细胞凋亡,探讨细胞凋亡在糖尿病心肌病变中的作用及意义。方法建立糖尿病大鼠模型,分别饲养12周及24周,采用核苷酸末端转移酶介导DUTP缺口翻译法(TUNEL)及透射电子显微镜技术,检测糖尿病大鼠心肌病变过程中左室心肌的凋亡细胞。结果STZ糖尿病大鼠12周时出现心功能异常并可见凋亡的心肌细胞,随病变的加重,24周时凋亡细胞数目明显增多;透射电镜检查,糖尿病大鼠左室心肌组织中亦可见凋亡的心肌细胞(具有凋亡形态学特征)。结论糖尿病心肌病变过程中存在心肌细胞凋亡且有加速趋势,心肌细胞凋亡可能是导致糖尿病心脏功能障碍的重要原因之一。
Objective To observe the presence ofapoptosis and to explore its role in the process of diabetic myocardium pathological changes of rats. Methods Diabetic animal models were induced by streptozotocin on SD rats. At 12 and 24 weeks, the abnormal heart function of rats was confirmed by heart function detection. The dynamic changes of myocardial apoptosis in left venticle of diabetic rats were investigated using TdT-mediated dUTP nick end labeling(TUNEL) and transmission electron microscope. Results Diabetic rats exhibited abnomal heart fuction and myocyte apoptosis at 12 weeks and aggravated at 24 weeks. Electron microscopic features of myocardial apoptosis were identified in left ventricle. Conclusion Myocardial apoptosis exsists in diabetic rats and it may be one of the most important factors which caused the abnormal diabetic heart function.
