桃红四物汤加味对兔复发性口腔溃疡中内皮素-1的表达及与血瘀证的相关性研究

Effect of Taohong Siwu Decoction on ET-1 in Recurrent Oral Ulcer in Rabbit and Relationship with Blood-stasis Syndrome

目的探讨桃红四物汤加味对兔复发性口腔溃疡(ROU)中内皮素—1(ET—1)的表达分布和表达强度以及这种表达与溃疡发生的关系,ET—1在ROU发病中与中医血瘀证的相关性及ROU发病的相关机制。方法采用免疫组织化学法造成兔口腔溃疡模型,分组用免疫组织化学法检测ROU口腔黏膜中ET—1的表达分布和表达强度以及血液流变学的改变情况。结果桃红四物汤组、金施尔康组和模型组中ET—1的含量均高于正常对照组(P<0.01),但中、西药两治疗组ET—1含量明显低于模型组(P<0.05);桃红四物汤组明显低于金施尔康组(P<0.05);血液流变学的改变,模型组明显高于其余3组(P<0.01);桃红四物汤可以改善血液流变学,延长溃疡总间歇时间及减少总溃疡个数,与金施尔康组比较有显著性差异(P<0.05)。结论内源性ET—1水平异常升高与ROU发病关系密切。桃红四物汤加味可降低ET—1含量的异常表达强度,改善血液流变学,延长溃疡发作总间歇时间,减少总溃疡个数。ET—1在ROU血瘀证的形成中有着重要的作用。

Objective To investigate the expression of distribution and intensity of Endothelin- 1 （ ET- 1 ） in recurrent oral ulcer （ROU）, and the expression related to occurence of the ulcer correlation of ET-1 and syndrome of blood stasis in occurence of ROU. Method The oral ulcer models of rabbit were made with immtmohistochemical method, The expression distribution and intensity of ET- 1 and blood-activating fromulas were tested with irnmunohistochemical method in 4 groups,and changes of blood rheology in oral mucosa were observed. Result Compared with normal group, the expression intensity of ET- 1 in therapy group and model group were significantly higher（ P 〈 0.01 ）, but in therapy group, the expression intensity of E-q＇- 1 was significantly lower than that in model group （ P 〈 0.05 ） ; Compared with westem medicine control group, the expression intensity of ET- 1 in TCM therapy group was lower（ P 〈 0.05）. The hemorrheology indexes in 4 groups were significantly lower（ P 〈 0.01 ）. Taohong Siwu decoction can improve the hemorrheology indexes, prolong the total ulcer interion time and reduce total ulcer number, and the effect is superior to the western medicine control group （ P 〈 0.05）. Conclusion The abnormal increase of ET- 1 is related to ROU. Taohong Siwu decoction can decrease the abnormal expression intensity of ET- 1, improve the hemorrheology indexes, prolong the total ulcer interim time and total ulcer number,Which showed that ET-1 plays an important rule in the formation of blood-stasis syndrome.
