摘要 目的:利用蛋白质连接法合成吗啡人工全抗原。方法:混合酸酐法和碳二亚胺连接法,用BSA及OVA蛋白做载体蛋白,利用紫外光谱扫描及蛋白质电泳两种方法判断连接是否成功。结果:连接成功,连接产物分子量增加,且每分子牛血清白蛋白连接的吗啡分子数为40-48个,每分子卵清白蛋白(OVA)连接的吗啡分子数为13-16个。结论:此两种方法能成功合成吗啡人工抗原,且该抗原可作为吗啡免疫用抗原以及吗啡免疫检测方法建立所用包被抗原。 Objective: To synthesize morphine antigen by conjugating morphine to protein. Methods: Morphine was conjugated to protein by mixed anhydride method and EDC method, and the cartier protein was BSA and OVA. The result was confirmed by UV scan method and protein electrophoresis method. Results: The conjunction was successful and the molecular weights of protein conjugates increased, having 40 - 48 residues linked to a molecule of BSA and 13 - 16 residues to a molecule of OVA. Conclusion. Morphine antigen can be synthesized successfully by the two methods mentioned above, Which can be used as immunity antigen and coated antigen in immunoassay for morphine.
