摘要
采用多孢分离技术,经驯化筛选获得一个适应松杉木屑栽培的猴头菌新菌株宁猴6号,比较了不同碳源、氮源、碳氮比、温度、含水量和pH条件下菌丝生长的情况。结果表明,宁猴6号菌丝生长的最佳碳源是淀粉,平均菌丝生长速度3.7mm.d-1,最佳氮源为麦麸,平均菌丝生长速度3.6mm.d-1,最佳碳氮比是30∶1;宁猴6号接种于培养基4中,置于10℃、15℃、20℃、25℃、30℃和35℃的恒温箱中,观察菌丝生长,结果表明,25℃最适宜,平均菌丝生长速度0.6cm.d-1;宁猴6号在培养基5中,含水量调节至500g.kg-1、550g.kg-1、600g.kg-1、650kg.kg-1、700g.kg-1、750g.kg-1、800g.kg-1和850g.kg-1,其结果表明,最适含水量为650g.kg-1,平均菌丝生长速度为0.6 cm.d;最适pH值为5,平均菌丝生长速度为0.5 cm.d-1。
In order to exploit new medium resource, a new strain of Ninghou 6 was selected by multi-spore isolation, which was domesticated to be suitable for the needle leaves powder. The different conditions have been compared in the experiment. The results showed that the optimal carbon resource was starch ,with the average growth speed of 3.7mm·d^-1 and the optimal nitrogen resource was wheat bran,with the average growth speed of 3.6mm·d^-1 for mycelium. The optimal C/N ratio was 30 : 1. Ninghou 6 was planted in No. 4 medium and placed in different temperatures,such as 10℃, 15℃,20℃,25 ℃, 30℃, 35℃. The results indicated that the optimal temperature was 25℃, with the average growth speed of 0. 6cm·d^-1. Ninghou 6 was cultivated in No. 5 media with different water contents,such as 500g·kg^-1 ,550g·kg^-1 , 600g·kg^-1 ,650g·kg^-1,700g·kg^-1 ,750g·kg^-1,800g·kg^-1,850g·kg^-1. The results suggested that the optimal water content was 650g·kg^-1 ,with the average growth speed of 0. 6cm·d^-1. Meanwhile,the optimal pH value of the medium was 5 ,with the average growth speed of 0. 5cm·d^-1 for mycelium.
