去葡萄糖竹节参皂苷Ⅳa对缺氧复氧心肌细胞损伤的保护作用 被引量:10

摘要 目的研究去葡萄糖竹节参皂苷Ⅳa(DCⅣa)是否为龙牙木匆心木总皂苷抗心肌缺血的有效成分之一。方法取体外培养的新生大鼠心肌细胞建立缺氧6 h复氧3 h心肌细胞损伤模型,并于缺氧及复氧开始时分别加入终浓度为2.5,1.0和0.1 mg.L-1的DCⅣa。检测细胞培养上清液中乳酸脱氢酶(LDH)和肌酸激酶(CK)漏出量,丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性。MTT法检测心肌细胞存活率。结果DCⅣa(2.5,1.0 mg.L-1)显著减少缺氧及复氧后LDH和CK漏出量,降低MDA含量,提高SOD活性,提高心肌细胞存活率。结论DCⅣa为龙牙木匆心木总皂苷抗心肌缺血的有效成分之一,可减轻缺氧复氧对心肌细胞的损伤,对心肌细胞具有直接的保护作用。 AIM To study whether deglucosechikusetsu saponin Ⅳa (DCIVa) is one of the effective components of aralosides to exert its protective action against myocardial ischemia. METHODS An anoxia (6 h)-reoxygenation (3 h) model of cultured myocardial ceils of neonatal Wistar rats was established, and DC Ⅳa was added into the culture medium with a final concentration 2.5, 1.0, 0.1 mg·L^-1 before anoxia and again before reoxygenation. Lactate dehydrogenase (LDH) and creatine kinase (CK) release, malondialdehyde (MDA) content and superoxide dismutase (SOD) activity in the culture medium were assayed. Myocardial ceils survival rate was measured by MTr assay. RESULTS DCⅣ a (2.5, 1.0 mg·L^-1) restrained evidently the LDH and CK release, decreased MDA content, enhanced SOD activity and increased myocardial cells survival rate. CONCLUSION DC Ⅳ a has direct protection for the cultured myocardial cells injured by anoxia and reoxygenation, it is one of the important ingredients of aralosides to exert its protective action against myocardial ischemia.
