摘要 目的:建立同时测定血浆中茶碱、苯巴比妥和卡马西平浓度的方法。方法:采用RP-HPLC,色谱柱:ZORBAX SB-C18 (4.6 mm×250 mm,5 μm),柱温:30℃,内标:乙酰苯胺,流动相:甲醇-水(52:48),流速:1 ml·min-1,检测波长:254 nm。结果:茶碱、苯巴比妥和卡马西平的线性范围分别为:2.6-41.6 μg·ml-1(r=0.999 1),4.97-79.52 μg·ml-1(r=0.999 2), 1.31-20.96 μg·ml-1(r=0.999 7)。平均回收率分别为99.3%,99.4%,99.9%;日内及日间RSD均小于5%。结论:该法简单快捷,灵敏度高,结果准确,避免联合用药的影响,适用于临床血药浓度监测。 Objective: To establish a method for simultaneous determination the concentration of theophilline, phenobartial, and carbamazepine in human plasma by RP-HPLC. Method: The ZORBAX SB -C18(4.6 mm×250 mm, 5μm)was used, the column temperature at 30℃, internal standard was acetanilide, the mobile phase was methanol-water(52:48 ), the flow rate was 1 ml·min^-1, detection wavelength was at 254 nm. Result: The linear ranges of theophilline, phenobartial, and carbamazepine were at of 2.6-41.6μg·ml^-1 (r =0.999 1) ; 4.97-79.52 g·ml^-1 (r =0.999 2) ; 1.31-20.96g·ml^-1 (r =0.999 7) respectively. The average recoveries of theophilline, phenobartial,and carbamazepine were 99.3% ,99.4% ,99.9% respectively. The RSD of within-day and betweendays were all less than 5 %. Conclusion: This method is simple, rapid, sensitive, accurate, It suitable for the clinical monitoring the blood concentrations.
