摘要 目的探讨多克隆抗体直接免疫荧光法检测白色念珠菌的最佳反应条件及临床应用意义。方法观察不同抗体稀释度(1:10~1:2 560),不同孵育时间(15、30、45、60、90、120 min),不同封闭剂(1%、10%牛血清白蛋白及10%小牛血清)和不同菌数对实验效果的影响。结果以抗体稀释度1:40、37℃孵育45 min、封闭剂1%或10%牛血清白蛋白以及菌数1.0×106个/ml和5.0×106个/ml为最佳反应条件。结论直接免疫荧光法检测白色念珠菌具有一定的优点和临床应用价值。 Objective: A preliminary research to detect candida albicans by direct immunofluorescence assay (DFA) was performed and its significance of clinical application was discussed in this paper. Methods: The suspensions of candida albicans at a series of concentrations were added on the slides and fixed by acetone. The slides were blocked by 1% bovine serum albumin(BSA) or 10% BSA or 10% newborn calf serum(NCS) respectively, incubated with different dilutions of polyclonal goat anti-Ca IgG (1 : 20~1 : 2560) for different incubation time (15, 30, 45, 60, 90, 120 min). Results: The optimal result was achieved under the following conditions: blocking solution of 1% or 10% BSA, the dilution 1 : 40 of polyclonal antibody, incubation at 37℃ for 45 min and the concentration of candida alibancans at 1.0× 10^6/ml and 5.0× 10^6/ml. Conclusion: DFA could be a useful method to detect candida albicans in clinical application.
