摘要
研究水酶法提取玉米胚芽油热处理及酶解工艺参数。酶解过程中,同时加入2.0%蛋白酶和1.5%纤维素酶,程序升温(40℃2h-50℃8h),两种酶协同作用,能加快细胞壁降解及油脂游离,缩短酶解反应时间,反应10h,酶解过程油得率达到90%以上。运用纳滤技术回收玉米胚芽浸泡液和酶解液中可溶性蛋白和肽。纳滤过程对酶解液和浸泡液浓缩效果显著,但脱盐和脱酸效果不理想。高效液相色谱(HPLC)图谱表明,纳滤前后,酶解液和浸泡液中蛋白质相对分子量分布发生变化。
In this paper, aqueous enzymatic extraction of corn germ oil and protein recovery by nanofiltration were studied. During enzymatic process, 2.0% proteinase and 1.5% cellulose were added at the same time, temperature was adjusted to 40℃ for 2h then to 50℃ for 8h. The oil output was more than 90% of the total oil in the corn germ. Nanofiltration technique was used to obtain soluble protein and peptide. The effect of concentration by nanofiltration was very remarkable, but the effect of salt and acid removal was unsatisfactory. The HPLC analysis showed that molecular weight distribution of hydrolysates and steeping water changed after nanofiltration.
