Hira基因的过量表达对果蝇早期胚胎发育的影响 被引量:5

摘要 H ir/H ira基因产物是组蛋白基因表达的一种负调节因子,其在果蝇发育过程中的作用还没有得到确认.本研究将果蝇H ira基因(dH ira)的cDNA克隆到载体UA SP中,运用UA S-G a l4系统使其在果蝇早期胚胎中大量表达.结果发现在胚胎发育早期,无论是在头部还是在全胚胎过量表达H ira,都引起果蝇胚胎大量死亡,而且随着转基因拷贝数的增加,胚胎的死亡率也显著增加,说明H ira过量表达对果蝇胚胎发育产生严重影响.由于H ira基因产物与核小体的组装、染色质结构等的调节有关,因此推测H ira过量表达可能是通过对组蛋白的抑制对果蝇胚胎发育造成影响的. HIR/HIRA, encoded by Hir/Hira (histone regulation) genes, were identified as negative regulators of histone gene expression. The function of the homolog ot hira gene in Drosophila (Hira) during the embryogenesis of Drosophila melanogaster has not been elucidated accurately. The Hira gene was cloned into UASP vector and overexpressed by UAS-Gal4 system in the early embryogenesis. The overexpression of Hira caused significantly increased lethality of embryos, which suggest that Hira gene may have important functions in embryogenesis of Drosophila. Since the products of Hira genes are involved in histone, the overexpression of Hira may affect Drosophila embryogenesis through the pathway related to histone.
