摘要
通过测定油松种子超低温贮藏后的电导率、脱氢酶活性、发芽率和发芽指数,对油松种子长期超低温贮藏的可行性进行研究。结果表明:超低温可以应用于油松种子的贮藏;冷冻方式和解冻方式是影响油松种子超低温贮藏的重要因素;防冻保护剂可有效减少种子在超低温贮藏时受到的损伤,二甲基亚砜的保护作用较为明显。试验的最佳处理组合为:10%二甲基亚砜+快速冷冻+慢速解冻。
By investigating conductivity, dehydrogenase, germination percentage and germination index of cryopreserved Chinese pine seeds, feasibility of long-term storage of the seeds was studied. The results showed that:(1)the freezing and thawing was the main factor affecting the cryopreservation of the seeds; (2) cryoprotectant reduced the damage of the seeds cryopreserved,DMSO's function was comparatively obvious; (3)the cryopreservation can be applied to store the Chinese pine seeds, and the optimal treatment combination was 10 % DMSO + quick freezing + slow thawing.
