中风后遗症肾虚痰瘀大鼠模型的建立

Establishing of rat model with stroke sequelae shenxutanyu

目的建立一种符合中医理论的中风后遗症“肾虚痰瘀”大鼠模型。方法在复制大鼠大脑中动脉梗死模型的基础上,根据中医“恐伤肾”及“过食肥腻滋生痰浊”的理论,采用猫吓鼠法及高脂饮食法制作"肾虚痰瘀"病理模型。结果模型组和模型对照组大鼠均出现了偏瘫症状,同时模型组大鼠还出现了中医肾虚痰瘀的证候特点:毛发卷曲黯淡无泽、精神萎靡、反应迟钝、嗜睡、表情惊恐,饮水减少,二便失禁(粒次数增多、粪便颗粒变小);生化指标也出现了相应的改变:模型组血脂含量及血液流变学指标均显著高于模型对照组(P<0.01和0.05),模型组脑组织及血浆MDA含量[脑组织MDA(165.27±7.61)nmol/g;血浆MDA(155.27±7.53)nmol/g]也显著高于模型对照组[脑组织MDA(153.94±5.73)nmol/g;血浆MDA(153.89±5.67)nmol/g](P<0.05),模型组脑组织及红细胞SOD活性[脑组织SOD(72.44±0.55)Nu/mg;红细胞SOD(60.44±0.55)抑制%]均显著低于模型对照组[脑组织SOD(79.97±0.43)Nu/mg;红细胞SOD(64.97±0.34)抑制%](P<0.05)。结论本方法可成功制作中风后遗症“肾虚痰瘀”大鼠模型,该模型可作为研究中风后遗症的实验基础。

Objective To establish a rat model with stroke sequelae “shenxutanyu” according with traditional chinese medicine theory. Method On the base of copying cerebral middle artery infarction model of rat, and according with traditional chinese medicine theory of “ fear hurt kidney” and “greasy excess cause sputum thicky”, pathological model of “shenxutanyu” was made by using the cat to frighten the rat and high fat dietary. Results Hemiplegia symptom is presented both in model rat and control rat. Meanwhile, the model rat also showed characteristic of “shenxutanyu” syndrome： convoluted and gray hairs, depressed, dullness, somnolence, panic, and reduced hydroposia, urinary and fecal incontinence （frenqency and small faeces） ; microindex showed corresponding change： the content of blood - fat and index of blood rheology in model group are all higher than control group （ P〈0.01 and 0.05）, while MDA content in brain tissue and blood plasma of model group is higher than control group, in contrast, SOD activity of brain tissue and red cell in model group is lower than control group （ P 〈 0.05）. Conclusion Rat model with stroke sequelae “shenxutanyu” can made successfully with this method, and the model can be taken as experimental basic for studying stroke sequelae.
