摘要
目的探讨用反相高效液相色谱法测定家兔血清中茶碱的浓度.方法采用C18柱,流动相为甲醇-水(35:65),检测波长275nm,柱温为室温,测定茶碱的血药浓度.血清中的茶碱用氯仿-异丙醇(95:5)提取.结果茶碱浓度在14.93~51.6μg/ml范围内与峰面积线性关系良好,线性方程为Y=6441.5x-76922,r=0.9947.结论用本法对6只家兔进行了血药浓度监测.该法取样量少,快速、灵敏、准确,可用于常规茶碱血药浓度的监测和进行药代动力学研究.
Objective To determine theophylline in serum by HPLC. Methods external standard method, The separation was performed on C18 (1.5L × 4.6) column with methanol - water(35:65) as mobile phase. The flow rate was 1.0mL/min. The wavelength for detection was 275nm. Theophylline was extracted from serum using a mixture of trichloromethane:isopropanol (95:5,V/V). Results A good linear relationship was obtained between the peak area and the concentration of theophylline from 4.31 to 43.08μg/ml, r = 0. 9537. The average recoveries of the method was 32.00 % and the average extraction recovery was 103.00 %. Conclusion This method is accuate,sensitive, rapid and simple. It can be used as a roution method by TDM.
