摘要
采用传统的饱和酚提取法和上海生物工程公司的UN IQ-10柱式基因组DNA抽提试剂盒法对大弹涂鱼的总DNA进行了提取。琼脂糖凝胶电泳结果表明,得到的DNA片段分子量在21 kb以上;通过PCR扩增实验,证明了用两种方法提取的DNA均可直接用于随机扩增的DNA多态性(RAPD)遗传标记;试剂盒法具有简单、快速、高效的优点,是一种值得推荐的DNA提取方法。
Using the traditional saturated-phenol distillation method and the kitted method, in which the UNIQ-10 kit is the product of Shanghai Biology Engineer Company, the genomic DNA of Boleophthalmus pectinirostris was extracted. The results of agarose electrophoresis showed that the fragments of genomic DNA samples were larger than 21 kb. The PCR results showed that DNA extracted by the two methods could be used directly for RAPD analysis. Comparing with the two methods, the kitted method is simple, fast and high-efficiency, so that it is worthwhile to recommend the kitted method.
